dc.description CONDICIONAMIENTO DE PUBLICACION DE PROYECTO. La influenza aviar H5N1 representa una amenaza critica para la seguridad alimentaria y la biodiversidad en Ecuador, evidenciando la necesidad de herramientas profilácticas mas eficaces y de fácil administración. El presente trabajo diseña in silico una plataforma vacunal probiótica modular utilizando lactococcus lactis modificado genéticamente para la expresión de antígenos virales conservados. Metodológicamente , se seleccionaron secuencias consenso de neuraminidasa, hemaglutinina y el ectodominio de matriz 2, mediante filogenia molecular. Se construyeron vectores de expresión optimizados y se valido el plegamiento proteico tridimensional empleando AlphaFold y dinamica molecular. Adicionalmente, se implemento un modelo matemático determinista para simular y optimizar la cinética de producción del sistema inducible NICE. Los resultados demostraron una alta conservación genética del virus, en la region, justificando un enfoque vacunal regional. Las predicciones estructurales ratificaron la estabilidad y correcta exposición de los epítopos quiméricos en la superficie bacteriana. Asimismo, el modelo matemático identifico una dosis operativa de nisina significativamente menor a la estándar, maximizando la eficiencia del bioproceso. Se concluye que la plataforma probiótica propuesta es teóricamente viable , segura y adaptable, constituyendo una solución prometedora y costo-efectiva para mitigar el impacto de enfermedades virales emergentes en el sector avícola nacional. es_EC
dc.description.abstract CONDITION FOR PROJECT PUBLICATION. Avian influenza H5N1 poses a critical threat to food security and biodiversity in Ecuador, highlighting the need for more effective and easily administered prophylactic tools. This study presents the in silico design of a modular probiotic vaccine platform using genetically modified Lactococcus lactis for the expression of conserved viral antigens. Methodologically, consensus sequences of neuraminidase, hemagglutinin, and the matrix 2 ectodomain (M2e) were selected through molecular phylogenetic analysis. Optimized expression vectors were constructed, and three-dimensional protein folding was validated using AlphaFold and molecular dynamics simulations. Additionally, a deterministic mathematical model was implemented to simulate and optimize the production kinetics of the Nisin-Controlled Expression (NICE) inducible system. The results demonstrated a high degree of viral genetic conservation within the region, supporting a regional vaccine approach. Structural predictions confirmed the stability and proper surface exposure of the chimeric epitopes on the bacterial surface. Furthermore, the mathematical model identified an operational nisin dose significantly lower than the conventional standard, thereby maximizing bioprocess efficiency. It is concluded that the proposed probiotic platform is theoretically feasible, safe, and adaptable, representing a promising and cost-effective strategy to mitigate the impact of emerging viral diseases in Ecuador's poultry sector.
